Mazzara is an Italian-American. He was born in Manhattan and brought up in Queens, New York. He went St. Francis Preparatory School where he graduated in 1985. He later proceeded to Ney York University and earned a masters in English before working as a hospital administrator in New York. He left New York for Los Angeles in 1998.

In 1998, Glen Mazzara moved from New York to Los Angeles to pursue a career as a writer in 1998. He became a story writer and editor for the fourth and fifth seasons of Nash Bridges. Glen Mazzara then joined The Shield, a drama series, as an executive story editor and writer for its first season in 2002. Mazzara was promoted steadily to producer in the second season while still writing, then to supervising producer for the third season, then co-executive producer for the fourth season and to executive producer for the fifth season. However, in 2006, he left midway through the sixth season. Later in the same year, 2006, he was the executive producer of the short-lived series Standoff. Mazzara was the co-executive producer on the first season of NBC drama Life.

He left Life to create a new series, Crash, as the executive producer. Crash was based on the Oscar-winning film of the same name on which he served as head writer and Director. Mazzara left after one season on Crash but was credited as a consulting producer for the second season premiere. Mazzara joined the medical drama series Hawthorne for two seasons as executive producer and writer in 2009. He then wrote his first of several episodes of the AMC series The Walking Dead, in 2010.
